Combination of 6-Benzylaminopurine and Thidiazuron Promotes Highly Efficient Shoot Regeneration from Cotyledonary Node of Mature Peanut (Arachis hypogaea L.) Cultivars

Efficient in vitro plantlet regeneration is an important step to successfully transform genes for the improvement of agronomic traits. A combination 6-benzylaminopurine (BAP) and thidiazuron (TDZ) plant growth regulators was applied evaluate shoot capacity whereas α-naphthalene acetic acid (NAA) with (BAP), 2, 4-dichlorophenoxyacetic (2, 4-D) were tested optimize root induction two peanut cultivars. The result showed (BAP TDZ) found be effective promoting shoot. highest frequency (93%) obtained on a medium supplemented 4 mg/L BAP 0.5 TDZ while average (87%) achieved containing combinations 2 1 TDZ. shooting rate increased both cultivars as concentrations decreased. rooting 3.5 NAA 2.5 concentration auxin cytokinin ratio increased. maximum (83%) MS 0.3 4-D 0.2 cultivar N3. indicated that much better than rate. Thus, protocol developed genotype independent tissue culture.
